Buccutite 交聯技術提供了最方便、最有效的交聯方法,以高共軛產率連接兩個生物分子。該方法使用一對交聯劑:Buccutite MTA 和 Buccutite FOL。 MTA 被添加到一個分子中,而 FOL 被添加到另一個分子中。通過在中性條件下混合 Molecule-1-Buccutite MTA 和 Molecule-2-Buccutite FOL 來引發交聯反應。Buccutite MTA 馬來酰亞胺 (MTAM) 的使用方式與廣泛用于交聯蛋白質的 SMCC 相同。MTAM 的一端(通過馬來酰亞胺)與還原抗體(通過 TCEP 或 DTT)中發現的半胱氨酸的硫醇 (-SH) 反應。 SMCC 交聯需要高濃度的蛋白質。此外,SMCC 修飾的蛋白質極不穩定,通常會自我反應,因為蛋白質通常同時含有胺和硫醇基團,這會導致大量的同交聯。 Buccutite 交聯反應在極其溫和和中性的條件下發生,無需任何催化劑,它比SMCC更加穩定高效。
