產品介紹

碘化丙啶(PI)屬于與溴化乙錠相同的化學類別。與溴化乙錠的情況一樣,其與熒光結合后的熒光增強20-30倍。熒光激發最大值紅移30-40nm,熒光發射最大值藍移15nm左右。PI也與DAPI和吖啶橙結合RNA。PI通常用于鑒定細胞群中的死細胞,并用作多色熒光技術中的復染劑。它還可用于區分壞死細胞,凋亡細胞和正常細胞。它適用于熒光顯微鏡,流式細胞儀和熒光測定法。

染色細胞示例

        PI染色通常在所有染色后進行。它只能染死細胞。以下程序適用于大多數細胞類型。生長培養基,細胞密度,其他細胞類型的存在和其他因素可能影響染色。玻璃器皿上的殘留洗滌劑也可能影響許多生物染色,導致在有/沒有細胞存在的溶液中出現明亮染色的物質。

1)制備1-10mM DMSO儲備溶液。如果儲存在-20°C,DMSO儲備液可以保存6個月。

2)使用適合您樣品的固定方案。

3)通過離心沉淀細胞,并將細胞重懸于緩沖鹽溶液或培養基中,在pH 7.4下進行最佳染料染色。培養物中的貼壁細胞可以在蓋玻片上或細胞培養孔中原位染色。

4)使用0.5和5μM之間的濃度添加PI染色,并將其孵育15至60分鐘。

注意:在最初的實驗中,最好在整個建議的范圍內嘗試幾種染料濃度,以確定產生最佳染色的濃度。
