簡要概述

4-甲基傘形酮磷酸二鈉鹽三水 MUP二鈉鹽是美國AAT Bioquest生產的MUP,MUP二鈉鹽是磷酸酶的熒光底物,它廣泛地用于溶液中磷酸酶的檢測。然而這種磷酸酶底物并不適用于活細胞或者連續性分析,因為MU(4-甲基傘形酮),酶促反應產物,只有在PH>10時才發出最大熒光。因此用MUP檢測酸性范圍內磷酸酶仍然是個難題,例如檢測酸性磷酸酶。AAT Bioquest很高興為大家提供MUP Plus,它與MUP底物相聯系克服了其pH的限制性。百螢生物是AAT Bioquest的中國代理商,為您提供最優質的4-甲基傘形酮磷酸二鈉鹽三水 MUP二鈉鹽。

產品說明書

實驗方案

1.準備工作:

1.1在升值溶劑中制備2至10 mM的儲備溶液(參見表)。 應立即使用原液。 任何未使用的溶液需要等分并在<-20℃冷凍。

注1:不要使用DMSO,ETOH或METH制備SunRed™磷酸鹽(目錄號11629)的儲備溶液,因為它會顯著增加分析背景。

注2:避免反復凍融循環,避免光照。

1.2制備2X磷酸鹽工作溶液:在實驗當天,將底物溶解在升值溶劑中或在室溫下解凍等份的儲備溶液(來自步驟1.1)。 在100 mM Tris緩沖液或您選擇的緩沖液(pH 8至9)(非磷酸鹽緩沖液)中制備10至50μM的2X工作溶液。

 

2.在上清液中進行磷酸酶測定:

2.1將50μL2X磷酸鹽工作溶液(來自步驟1.2)加入磷酸酶標準品,空白對照和測試樣品的每個孔中,使總磷酸酶測定體積為100μL/孔。

注意:對于384孔板,每孔加入25μL樣品和25μL2X磷酸鹽工作溶液。

2.2在所需溫度下孵育反應30至120分鐘,避光。

2.3用熒光板讀數器監測適當濾光片組(見表)的熒光增加。

2.4空白孔中的熒光(僅含測定緩沖液)用作對照,并從具有磷酸酶反應的那些孔的值中減去。
