標準操作規程(標記50μg蛋白質)

      將所有組分加熱并在打開前將樣品瓶短暫離心,并在開始結合前立即準備所需的溶液。 以下SOP是標記抗HDAC IgG抗體的實例。

 

1.準備蛋白質溶液(溶液A):

為了標記50g蛋白質(假設目標蛋白質濃度為1mg / mL),將5L(總反應體積的10%)的反應緩沖液(組分B)與50L目標蛋白質溶液混合。

注1.如果您的蛋白質濃度不同,請相應調整蛋白質體積,使~50μg蛋白質可用于標記反應。

注2:為了標記100g蛋白質(假設目標蛋白質濃度為1mg / mL),將10μL(總反應體積的10%)反應緩沖液(組分B)與100μL目標蛋白質溶液混合。

注3:蛋白質應溶解于pH7.2-7.4的1X磷酸鹽緩沖鹽水(PBS)中;如果蛋白質溶解在甘氨酸緩沖液中,必須用1X PBS(pH 7.2-7.4)透析,或使用Amicon Ultra-0.5,Ultracel-10膜,10 kDa(來自Millipore的cat#UFC501008)去除游離胺或銨鹽(如硫酸銨和乙酸銨)廣泛用于蛋白質沉淀。

注4:用牛血清白蛋白(BSA)或明膠穩定的不純抗體或抗體不能很好地標記。

注5:如果蛋白質濃度小于1 mg / mL,則結合效率顯著降低。為獲得最佳標記效率,建議最終蛋白質濃度范圍為1-2 mg / mL。

 

2.運行結合反應:

2.1將蛋白質溶液(溶液A)加入一瓶標記染料(組分A)中,通過反復移液幾次將其混合均勻,或將小瓶渦旋幾秒鐘。

注意:使用標記染料的兩個小瓶(組分A)通過將100μg蛋白質分成2x50μg蛋白質并使每個50μg蛋白質與一小瓶標記染料反應來標記100μg蛋白質。 將兩個小瓶合并用于下一步。

2.2將綴合反應混合物在室溫下保持30-60分鐘。

注意:如果需要,可以旋轉或搖動綴合反應混合物更長的時間。

 

3.停止共軛反應:

3.1添加5 L(對于50μg蛋白質)或10L(對于100μg蛋白質),其為TQ染色猝滅緩沖液(組分C)的總反應體積的10%到綴合反應混合物中(來自步驟2.2),將它們混合 好。

3.2在室溫下孵育10分鐘。

3.3標記的蛋白質(抗體)現在可以使用了。
